چکیده

a 2 -induced Ca 2 release (CICR) plays an important role in the generation of cytosolic Ca 2 signals in many cell types. However, it is inherently difficult to distinguish experimentally between the contributions of messenger-induced Ca 2 release and CICR. We have directly tested the CICR sensitivity of different regions of intact pancreatic acinar cells using local uncaging of caged Ca 2 . In the apical region, local uncaging of Ca 2 was able to trigger a CICR wave, which propagated toward the base. CICR could not be triggered in the basal region, despite the known presence of ryanodine receptors. The triggering of CICR C from the apical region was inhibited by a pharmacological block of ryanodine or inositol trisphosphate receptors, indicating that global signals require coordinated Ca 2

Ca2+-Induced Ca2+ Release through Localized Ca2+ Uncaging in Smooth Muscle

Ca(2+)-induced Ca(2+) release (CICR) from the sarcoplasmic reticulum (SR) occurs in smooth muscle as spontaneous SR Ca(2+) release or Ca(2+) sparks and, in some spiking tissues, as Ca(2+) release that is triggered by the activation of sarcolemmal Ca(2+) channels. Both processes display spatial localization in that release occurs at a higher frequency at specific subcellular regions. We have use...

Calcium Wave Propagation in Pancreatic Acinar Cells

In pancreatic acinar cells, inositol 1,4,5-trisphosphate (InsP(3))-dependent cytosolic calcium ([Ca(2+)](i)) increases resulting from agonist stimulation are initiated in an apical "trigger zone," where the vast majority of InsP(3) receptors (InsP(3)R) are localized. At threshold stimulation, [Ca(2+)](i) signals are confined to this region, whereas at concentrations of agonists that optimally e...

Synaptically activated Ca2+ waves and NMDA spikes locally suppress voltage-dependent Ca2+ signalling in rat pyramidal cell dendrites.

Postsynaptic [Ca(2+)](i) changes contribute to several kinds of plasticity in pyramidal neurons. We examined the effects of synaptically activated Ca(2+) waves and NMDA spikes on subsequent Ca(2+) signalling in CA1 pyramidal cell dendrites in hippocampal slices. Tetanic synaptic stimulation evoked a localized Ca(2+) wave in the primary apical dendrites. The [Ca(2+)](i) increase from a backpropa...

Analysis of two kinetically distinct components of transmitter release at a fast synapse of the mammalian central nervous system

6 Summary Summary Two kinetically distinct components of transmitter release have been observed in Ca 2+ uncaging experiments at a CNS synapse in rats, and both release components showed a steep dependence of release kinetics on [Ca 2+ ] i. These findings were based on simultaneous pre-and postsynaptic whole-cell voltage-clamp recordings from the calyx of Held synapse, combined with presynaptic...

Ca2+ sparks and puffs are generated and interact in rat hippocampal CA1 pyramidal neuron dendrites.

1,4,5-Inositol trisphosphate receptors (IP3Rs) and ryanodine receptors (RyRs) mediate release of Ca(2+) from internal stores in many neurons. The details of the spatial and temporal characteristics of these signals and their interactions in dendrites remain to be clarified. We found that localized Ca(2+) release events, with no associated change in membrane potential, occurred spontaneously in ...
